Promising Early Results for a New Parkinson’s Treatment: ION859
April 15, 2025
LeahJSEarly clinical trial results suggest that ION859, an investigational treatment being developed by Ionis Pharmaceuticals, was safe and well tolerated in people living with Parkinson’s disease.
Targeting LRRK2 — A Protein Linked to Parkinson’s
ION859 is designed to lower the levels of LRRK2, a protein that can become overactive in Parkinson’s disease. High LRRK2 activity is believed to contribute to nerve cell damage, even in people without a mutation in the LRRK2 gene.
How the Trial Worked
In the Phase 1 study, 82 people living with Parkinson’s received ION859 through spinal canal injections, either as a single dose or repeated monthly doses. Researchers measured both safety and whether the treatment reduced LRRK2 protein levels in the fluid surrounding the brain and spinal cord.
Encouraging Signs Beyond Safety
In addition to being well tolerated, ION859 reduced LRRK2 protein levels and other markers involved in clearing cellular waste — a process that can break down in Parkinson’s and contribute to disease progression. The strongest effects were seen at higher doses and the changes lasted for at least three months.
Looking Ahead
While this is only an early-stage study, these results offer hope that ION859 could one day help slow Parkinson’s progression by targeting an important disease pathway, not just managing symptoms. More research is planned to learn whether this approach can make a meaningful difference for people living with Parkinson’s.
