Promising New Findings on Solengepras for Early Parkinson’s Disease

Solengepras (CVN424), an oral therapy being developed by Cerevance, may offer a new option for people newly diagnosed with Parkinson’s disease. According to recent results from the Phase 2 ASCEND trial, solengepras helped ease both functional and nonmotor symptoms when used alone, before the start of standard Parkinson’s medications. A Closer Look at the Trial The ASCEND trial included 64 adults who had not yet started Parkinson’s treatment. Participants received either solengepras or a placebo daily for three months. The main goal was to assess how the treatment affected motor symptoms and daily functioning using patient and clinician assessments. While changes in motor symptoms were small and not statistically significant, people taking solengepras reported improvements in their ability to manage everyday activities. Support for Nonmotor Symptoms Many people living with Parkinson’s experience challenges that go beyond movement—such as sleep difficulties, changes in mood, or reduced motivation. The ASCEND trial found that solengepras may help with these nonmotor symptoms, based on self-reported outcomes. Fewer individuals on solengepras experienced nonmotor side effects compared to those on placebo. How Solengepras Works and Safety Profile Solengepras is a small molecule that reaches the brain and acts on a receptor called GPR6, located in a region important for voluntary movement. Rather than changing dopamine levels directly—as many current treatments do—solengepras offers a different approach that may reduce common side effects like involuntary movements. The treatment was generally well tolerated, with no serious side effects reported. Looking Ahead These findings add to earlier research showing that solengepras may also help when used alongside standard treatments like levodopa. It is now being studied further in the ongoing Phase 3 ARISE trial. While more research is needed, solengepras may represent a hopeful step toward more options for those navigating the early stages of Parkinson’s disease.
